A Phase 3 Study to Evaluate the Safety and Efficacy of KarXT + KarX-EC for the Treatment of Agitation Associated With Alzheimer's Disease (ADAGIO-2)

This research study, called ADAGIO-2, is studying a new treatment for agitation, which is when people with Alzheimer's disease feel restless, upset, or sometimes aggressive. The treatment being tested is a combination of medicines called KarXT and KarX-EC. The main goal is to find out if this treatment is safe for people to take and if it effectively helps to reduce these feelings of agitation. Some patients will receive the new treatment, while others will receive a placebo (a dummy pill with no active medicine). This type of study helps doctors understand if the new treatment works better than no treatment at all. It's for adults aged 55 to 90 who have been diagnosed with Alzheimer's and experience agitation.

What is this study about?

Imagine you or a loved one has Alzheimer's disease and sometimes feels very restless, upset, or even aggressive. This can be challenging for everyone involved. This study, called ADAGIO-2, is exploring a new medication combination, KarXT + KarX-EC, to see if it can help calm these feelings of agitation. The researchers want to discover if this treatment is both safe to use and effective in making people feel more settled.

This is a 'Phase 3' study, which means the treatment has already shown some promise in earlier tests. Now, it's being tested on a larger group of people to confirm its benefits and side effects. Some participants will receive the active treatment, while others will get a placebo, which looks identical but contains no medicine. This helps the researchers accurately compare the new treatment's effects against no active treatment.

The study aims to provide clearer information about whether KarXT + KarX-EC could become a helpful option for managing agitation in Alzheimer's. Finding better ways to manage agitation can significantly improve the daily lives of people with Alzheimer's and their families.

Key takeaways

  • This study tests a new treatment (KarXT + KarX-EC) for agitation in Alzheimer's.
  • It aims to find out if the treatment is safe and effective.
  • Participants will either receive the new treatment or a placebo.
  • A main caregiver's involvement is important for participants.
  • Taking part means regular visits and health checks over time.
  • You can stop participating at any point you wish.

Who may be eligible?

To join this study, people need to be between 55 and 90 years old and have a confirmed diagnosis of Alzheimer's disease. This diagnosis needs to be backed up by specific medical tests, such as special brain scans or blood/spinal fluid tests that look for signs of Alzheimer's.

Participants also need to be experiencing agitation, which means they feel restless, upset, or sometimes aggressive. This agitation must have started at least two weeks before the study begins and be significant enough to meet certain criteria set by the researchers. Their memory and thinking skills, checked by a simple test called the MMSE, should be within a specific range.

Finally, a key requirement is having a main caregiver who spends at least 10 hours a week with the participant. This caregiver must be willing to attend all study visits, help ensure the participant takes their medication correctly, and provide information about how the participant is doing throughout the study.

Quick self-check
  • Are you between 55 and 90 years old?
  • Do you have a diagnosis of Alzheimer's disease confirmed by specific medical tests?
  • Are you currently experiencing agitation (restlessness, being upset)?
  • Is there a family member or friend who acts as your main caregiver, spending at least 10 hours a week with you and willing to help with the study?

This is a guide only — the research team will confirm whether you can take part.

What does participation involve?

If you decide to take part, you'll have regular visits to the study clinic. These visits will involve various assessments, like answering questions about how you're feeling and your memory, and doctors will check your overall health. You will also be given either the study medication (KarXT + KarX-EC) or a placebo, which you'll need to take as directed. Your caregiver will play a vital role in helping with these visits and making sure the medication is taken correctly. The study will run for a specific period, and you'll have follow-up checks even after finishing the main treatment period to ensure your well-being. The total duration of participation will be explained to you in detail.

Potential risks and benefits

Participating in this study might offer the potential benefit of reducing agitation related to Alzheimer's disease. However, like all medicines, the study drug might have side effects, and some are unknown. You would be closely monitored for any reactions. It's important to remember that you are free to withdraw from the study at any time, for any reason, without it affecting your medical care.

Common questions

What is agitation in Alzheimer's disease?

Agitation means feeling restless, upset, or sometimes showing aggressive behaviours, which can happen to people with Alzheimer's disease.

What is KarXT + KarX-EC?

It's a new combination of medicines being tested to see if it can help reduce agitation in people with Alzheimer's disease.

What is a placebo?

A placebo is a 'dummy' pill that looks just like the study medicine but contains no active ingredients. It helps researchers compare the real treatment's effects.

Why is a caregiver needed for this study?

A caregiver is essential to help with study visits, make sure medication is taken as prescribed, and report on how the participant is doing, providing useful information to the research team.

Can I leave the study once I've joined?

Yes, you can choose to leave the study at any time, for any reason, and it will not affect your ongoing medical care.
