A Phase 2b, multicenter, randomized, double-blind study of safety and efficacy of TAK-755 (rADAMTS13) with minimal to no plasma exchange (PEX) in the treatment of immune-mediated thrombotic thrombocytopenic purpura (iTTP)

This research is investigating a new treatment called TAK-755 for a rare and serious blood disorder known as immune-mediated thrombotic thrombocytopenic purpura (iTTP). Currently, a treatment called plasma exchange (PEX) is often used, but it can be time-consuming and challenging. This study, called a Phase 2b trial, will see how safe and effective TAK-755 is for people with iTTP, specifically looking at whether it can allow patients to have less or even no plasma exchange. They will also look at how well patients recover, if the condition comes back, and any side effects. This research hopes to find a new, potentially easier way to help people with iTTP.

What is this study about?

This study is about a health condition called immune-mediated thrombotic thrombocytopenic purpura, or iTTP for short. iTTP is a very rare but serious blood disorder where tiny clots form in the blood vessels, which can cause problems in many organs. It happens because the body’s immune system mistakenly attacks a necessary protein in the blood. Currently, a main treatment for iTTP is plasma exchange (PEX), where a machine removes some of the patient's blood, filters out harmful parts, and returns it. While PEX is effective, it can be a difficult and time-consuming process for patients.

Researchers are looking into a new medicine called TAK-755 (also known as rADAMTS13). This medicine is designed to replace the missing protein that is faulty in people with iTTP. The main goal of this study is to find out if TAK-755 is safe and how well it works, especially when used with less or no plasma exchange. This is a 'Phase 2b' study, meaning it's still in the earlier stages of testing new medicines in people, but it's beyond the very first safety checks.

The hope is that TAK-755 could offer a new way to treat iTTP that might reduce the need for plasma exchange, making treatment easier for patients. The study will carefully watch for any side effects from TAK-755 and measure how quickly people get better, whether their iTTP returns, and if they need other treatments. This information will help doctors understand if TAK-755 could become a valuable new option for people living with iTTP.

Key takeaways

  • This study is testing a new medicine called TAK-755 for a rare blood condition called iTTP.
  • The main goal is to see if TAK-755 is safe and can reduce or remove the need for plasma exchange.
  • It's a Phase 2b study, meaning it's an earlier stage of testing in people.
  • Participants will be closely checked for how well the medicine works and any side effects.
  • Joining the study is voluntary, and you can leave at any time.

Who may be eligible?

To be able to join this study, participants generally need to be at least 18 years old. There won't be any restrictions based on whether you are male or female.

Beyond basic age and sex requirements, there will be more detailed health checks to make sure the study medicine is right for you and to keep you safe. For example, doctors will need to confirm you have active iTTP that has recently been diagnosed or has come back after treatment. They will also check if you have any other serious health conditions that might make it unsafe for you to take part in the study.

It’s important to remember that not everyone with iTTP will be able to join this study. The medical team will review your full health history and run specific tests to see if you meet all the criteria needed for this particular research.

Could this study suit you?

Answer these quick questions to see if you may be eligible. This is a guide only — the research team makes the final call.

  1. Are you 18 years old or older?
  2. Have you been diagnosed with iTTP, or has your iTTP recently returned?
  3. Are you able to attend regular hospital visits for treatment and check-ups?
  4. Are you willing to potentially receive a new, experimental medicine?
Answer every question to see your result.

What does participation involve?

If you join this study, you'll be given the study medicine, TAK-755, and potentially other treatments like Rituximab, according to a plan set by the study doctors. You will be closely monitored by the medical team throughout the study.

The study involves regular hospital visits for check-ups, blood tests, and to receive your treatment. These visits will help the doctors keep track of your condition, your response to the treatment, and watch for any side effects. You might also have physical examinations and other tests to see how the medicine is affecting your body and the iTTP.

The study will carefully record how often you need plasma exchange, if at all, and your overall recovery. The full duration of your participation in the study, including follow-up appointments after your treatment ends, will be explained by the study team.

Potential risks and benefits

Taking part in any medical study has potential benefits and risks. You might benefit from receiving a new medicine that could help manage your iTTP and potentially reduce your need for plasma exchange. However, as this is a new treatment, there might be side effects that doctors don't fully know about yet. There's also the chance the medicine might not work for you. The study team will carefully monitor you for any problems. You have the right to withdraw from the study at any time, for any reason, without it affecting your usual medical care.

Common questions

What is iTTP?

iTTP is a rare and serious blood condition where abnormal blood clots form, which can block blood flow to organs.

What is TAK-755?

TAK-755 is a new experimental medicine being tested in this study. It aims to replace a missing protein that is faulty in people with iTTP.

What is 'plasma exchange'?

Plasma exchange is a treatment for iTTP where a machine removes and cleans your blood by replacing certain parts, often used to remove harmful substances.

Will I still get my usual treatment in the study?

The study will test TAK-755, potentially alongside Rituximab. Your doctors will explain how this fits with or changes your current treatment plan.

What does a 'Phase 2b' study mean?

A Phase 2b study is a stage of research where doctors are learning more about how safe and effective a new medicine is for a specific condition.
